目的探讨一口钟不同溶剂提取物对大鼠佐剂性关节炎的治疗作用。方法采用不同有机溶剂对一口钟进行提取,得到各溶剂的提取物。将70只雄性Wistar大鼠随机分为正常组、模型组、石油醚组、三氯甲烷组、乙酸乙酯组、95%乙醇组和5%乙醇组各10只;正常组大鼠左后足跖皮内注射0.9%氯化钠注射液,每只0.1mL;其他各组大鼠左后足跖皮内注射弗氏完全佐剂,每只0.1mL。实验第10天,正常组和模型组给予0.9%氯化钠注射液灌胃,其他各组分别灌胃给予一口钟不同溶剂提取物,均0.5mL·(200g)-1,连续16d。检测各组大鼠足跖肿胀、脾脏指数、胸腺指数及血清一氧化氮含量。结果除一口钟石油醚提取物组外,与模型组比较,其他各提取物均能显著抑制大鼠左后足跖肿胀;一口钟不同溶剂提取物组大鼠胸腺指数、脾脏指数均显著增高,血清一氧化氮浓度显著降低。结论一口钟提取物对大鼠佐剂性关节炎有显著治疗作用,推测一口钟治疗佐剂性关节炎有效部位为脂溶性成分。
Objective To investigate the therapeutic effect of one or more different solvent extracts on adjuvant arthritis in rats. Methods One-minute extraction was performed using different organic solvents to obtain extracts of each solvent. 70 male Wistar rats were randomly divided into normal group, model group, petroleum ether group, trichloromethane group, ethyl acetate group, 95% ethanol group and 5% ethanol group; 10 rats in normal group; Intradermal injection of 0.9% sodium chloride injection, each 0.1mL; other groups of rats left tinea pedis injected Freund’s complete adjuvant, each 0.1mL. On the tenth day of the experiment, the normal group and the model group were given 0.9% sodium chloride injection gavage. The other groups were given intragastrically one minute of different solvent extracts, both 0.5 mL (200 g)-1 for 16 days. The paw swelling, spleen index, thymus index and serum nitric oxide level were measured in each group. Results Compared with the model group, all other extracts significantly inhibited the left hind paw edema in rats except for one oral petroleum ether extract group. The thymus index and spleen index of rats in different solvent extract groups were significantly increased. Serum nitric oxide levels were significantly reduced. Conclusion One-minute extract has a significant therapeutic effect on adjuvant arthritis in rats. It is speculated that a bell for the treatment of adjuvant arthritis is a fat-soluble component.
