为了研究铁皮石斛对环磷酰胺(cyclophosphamide,CY)所致免疫抑制模型小鼠血清中抗氧化酶活性及细胞因子含量的影响,试验选用100只雌性KM种小鼠随机分为5组,每组5个重复,每个重复4只。Ⅰ组为对照组,每天灌服0.3 m L纯化水;Ⅱ组为模型组,每天灌服0.3 m L纯化水;Ⅲ、Ⅳ、Ⅴ组为试验组,分别灌服1,2,3 g/kg铁皮石斛水煎剂。Ⅱ、Ⅲ、Ⅳ、Ⅴ组按体重100 mg/kg腹腔注射CY,隔日注射1次,连续15 d。结果表明:试验成功构建免疫抑制小鼠模型。与Ⅰ组(对照组)相比,Ⅱ组(模型组)、Ⅲ、Ⅳ、Ⅴ组小鼠血清中GSH-Px活性显著降低(P<0.05);各组间CAT活性差异均不显著(P>0.05);Ⅱ组(模型组)和Ⅲ、Ⅳ、Ⅴ组小鼠血清中IL-4、IFN-γ、IL-10含量均存在显著差异(P<0.05)。与Ⅱ组(模型组)相比,Ⅰ组(对照组)与和Ⅲ、Ⅳ、Ⅴ组小鼠血清中MDA含量显著降低(P<0.05);Ⅰ组(对照组)和Ⅴ组小鼠血清中SOD活性显著升高(P<0.05),Ⅲ、Ⅳ组小鼠血清中SOD活性有升高趋势(P>0.05);Ⅲ、Ⅳ、Ⅴ组小鼠血清中IL-2含量显著降低(P<0.05);Ⅰ、Ⅲ、Ⅳ及Ⅴ组小鼠血清中IL-4/IFN-γ比值差异显著(P<0.05)。说明铁皮石斛能通过调控CY构建的免疫抑制小鼠血液内细胞因子,从而对免疫抑制小鼠的血清抗氧化酶活性及Th1与Th2细胞发挥调节作用,使得血液细胞因子指标恢复正常,有效提高环磷酰胺所致的小鼠免疫调节机能,具有一定的免疫增强作用。
In order to study the effects of Dendrobium candidum on the activities of antioxidant enzymes and cytokines in serum of immunosuppressive mice induced by cyclophosphamide (CY), 100 female KM mice were randomly divided into 5 groups, each group Five replicates, four replicates each. The control group was fed with 0.3 m L of purified water daily. Group Ⅱ was treated with 0.3 m L of purified water daily. Groups Ⅲ, Ⅳ and Ⅴ were treated with 1, 2, 3 g / kg Dendrobium decoction. The results showed that the immunosuppressed mouse model was constructed successfully. Compared with group I (control group), GSH-Px activity in serum of group Ⅱ, model group, Ⅲ, Ⅳ and Ⅴ decreased significantly (P <0.05); CAT activity in all groups showed no significant difference (P > 0.05). The levels of IL-4, IFN-γ and IL-10 in group Ⅱ (model group) and Ⅲ, Ⅳ and Ⅴ groups were significantly different (P <0.05). Compared with group Ⅱ (model group), the content of MDA in serum of group Ⅰ (control group) and Ⅲ, Ⅳ, Ⅴ group was significantly decreased (P <0.05); the serum of group Ⅰ (control group) and groupⅤ (P <0.05). The activity of SOD in serum of mice in groups Ⅲ and Ⅳ increased (P> 0.05), and the levels of IL-2 in mice in groups Ⅲ, Ⅳ and Ⅴ decreased significantly (P < <0.05). The serum IL-4 / IFN-γ ratio in group Ⅰ, Ⅲ, Ⅳ and Ⅴ was significantly different (P <0.05). Dendrobium candidum could modulate the serum anti-oxidative enzyme activity of Th1 and Th2 cells in immunosuppressed mice by regulating the cytokines in the blood of immunosuppressed mice, so that the indexes of blood cytokines returned to normal, Phosphoramidic mice induced by immune function, with a certain degree of immune enhancement.
