目的 探讨血管内皮生长因子 (VEGF)对牛眼视网膜色素上皮 (RPE)细胞DNA合成的影响 ,并观察榄香烯注射液的抗增殖作用。方法 采用3H -TdR掺入法 ,在体外培养的牛眼RPE细胞中加入不同浓度的VEGF以及VEGF和榄香烯 ,应用液闪仪测定3H TdR掺入的脉冲数 (CPM)。结果 不同浓度的VEGF均使CPM值明显增加 ,且与VEGF浓度呈剂量依赖关系 ,不同浓度的榄香烯均使RPE细胞的CPM值明显下降 ,且与榄香烯浓度呈剂量依赖关系。结论 VEGF有促进RPE细胞的DNA合成的作用 ,榄香烯可抑制生长因子诱导的RPE细胞的DNA合成。
Objective To investigate the effect of vascular endothelial growth factor (VEGF) on the DNA synthesis in cultured bovine retinal pigment epithelium (RPE) cells and observe the antiproliferative effect of elemene injection. Methods 3H-TdR incorporation method was used to measure the number of incorporation of 3H-TdR (CPM) by adding different concentrations of VEGF, VEGF and elemene to bovine RPE cells cultured in vitro. Results Different concentrations of VEGF increased the CPM value in a dose-dependent manner. The concentrations of elemene decreased the CPM of RPE cells and the concentration of elemene in a dose-dependent manner. Conclusion VEGF can promote the DNA synthesis of RPE cells. Elemene can inhibit DNA synthesis induced by growth factor in RPE cells.
