目的 探讨血管内皮生长因子 (VEGF)血小板反应素 1(TSP 1)和微血管计数 (MVC)在眼肿瘤病人组织中的表达及其关系。方法 采用逆转录聚合酶链反应法 (RT PCR)和免疫组化S P法检测VEGF、TSP 1和MVC在 90例眼肿瘤病变组织中的表达 ,并用计算机图像分析系统对血管新生的强度进行定量分析。结果 肿瘤组织VEGF和MVC的表达同步增高 ;TSP 1mRNA的表达和MVC、VEGF mRNA有显著的负相关 (r =- 0 4 5 2 ,r=- 0 4 4 7,P均 <0 0 5 ) ;VEGFmRNA表达与MVC之间呈显著的正相关 (r =0 4 5 2 ,P <0 0 5 )。结论 眼肿瘤病人组织中VEGF及MVD的表达 ,可能与其血管化程度有关 ;TSP 1在肿瘤血管抑制方面发挥重要的作用。
Objective To investigate the expression and relationship of vascular endothelial growth factor (VEGF), thrombospondin 1 (TSP 1) and microvessel count (MVC) in ocular tumor tissues. Methods The expression of VEGF, TSP 1 and MVC in 90 cases of ocular neoplasia tissues were detected by reverse transcriptase polymerase chain reaction (RT PCR) and immunohistochemical SP method. The intensity of angiogenesis was quantitatively analyzed by computer image analysis system . Results The expression of VEGF and MVC in tumor tissue increased synchronously. The expression of TSP 1 mRNA was negatively correlated with MVC and VEGF mRNA (r = - 0 452, r = - 0 477, P 0 05). There was a significant positive correlation between VEGF mRNA expression and MVC (r = 0 452, P <0 05). Conclusion The expression of VEGF and MVD in ocular tumor tissue may be related to the degree of vascularization. TSP 1 plays an important role in tumor angiogenesis.
