试验以长白山野生白耙齿菌菌株为试验材料,进行ITS序列鉴定及人工驯化栽培试验。利用通用引物ITS1和ITS4进行ITS序列鉴定,同时利用农作物下脚料对白耙齿菌进行人工驯化栽培。原种培养料设置6个处理,栽培种培养料设置8个处理,通过ITS序列分析结果得出该菌株ITS序列与NCBI中登录号为JX290578.1、FJ462768.1、EU301643.1等9个白耙齿菌菌株相似性达100%,验证了其为白耙齿菌(Irpex lacteus);最佳原种培养料配方为木屑41.5%、豆秸41.5%、麦麸10%、黄豆粉5%、石膏1%、白糖1%,菌丝生长速度最快,为7.571 mm·d-1,且菌丝浓密,洁白,粗壮;最佳栽培种培养料配方为木屑54%、玉米芯29%、麦麸10%、黄豆粉5%、石膏1%、白糖1%,菌丝生长速度及长势显著高于其他处理。
In this study, the strain of white-streaked white tooth fungus from Changbai Mountain was used as experimental material to carry out ITS sequence identification and artificial domestication cultivation experiments. ITS1 and ITS4 were used for ITS sequence identification, at the same time, the use of crop waste on the white harrier bacteria artificial domestication cultivation. 6 strains of original culture material were set up and 8 varieties of culture material were set up. ITS ITS sequence and NCBI accession number of JX290578.1, FJ462768.1 and EU301643.1 were obtained by ITS sequence analysis. The results showed that the strains were Irpex lacteus with the similarity of 100%, the best cultivar was 41.5% wood chips, 41.5% soybean straw, 10% wheat bran and 5% Gypsum 1%, sugar 1%, mycelial growth fastest, 7.571 mm · d-1, and the mycelium thick, white, thick; best cultivars culture materials formula for the 54% sawdust, corn cob 29% 10% bran, 5% soy flour, gypsum 1%, 1% sugar, mycelial growth rate and growth was significantly higher than the other treatments.
