试验以无籽刺梨组培苗为材料,利用秋水仙素作为诱导剂,采用混培法,对二倍体组培苗进行诱导,比较不同预培养天数和不同浓度的秋水仙素对诱导效果的影响。结果表明,无籽刺梨茎段在分化培养基上预处理1d后,再接入含秋水仙素浓度为100mg/L的培养基中能得到较好的诱导结果,诱变率为7.8%。变异植株与对照相比,其叶形指数明显减少,叶色变深、叶柄变粗,保卫细胞和气孔明显增大。经根尖染色体制片计数后发现部分根尖细胞染色体为2n=4x=28,为四倍体。
