目的观察hLF对于人非小细胞肺癌H460细胞辐射敏感性的影响。方法用pBC1-hLF载体转染H460细胞,同时以hLF标准品处理为参照,细胞活力实验、克隆形成试验和流式检测细胞凋亡来观察hLF基因高表达对H460细胞生长特性和辐射敏感性的影响。结果hLF基因高表达可以显著抑制H460的生长(P<0.01),提高其对射线的敏感性,hLF处理组与pBC1-hLF转染组辐射增敏率分别为1.29和1.59。hLF可促进H460细胞自发和受照后的凋亡,降低克隆形成能力(P<0.01)。结论hLF基因高表达可以体外显著抑制人非小细胞肺癌H460细胞的生长,提高其辐射敏感性。
Objective To observe the effect of hLF on the radiosensitivity of human non-small cell lung cancer H460 cells. Methods H460 cells were transfected with pBC1-hLF vector and treated with hLF standard for reference. Cell viability assay, clonogenic assay and flow cytometry were used to observe the effect of hLF gene expression on the growth and radiation sensitivity of H460 cells influences. Results The high expression of hLF gene could significantly inhibit the growth of H460 cells (P <0.01) and enhance the sensitivity to radiation. The sensitivities of hLF-treated cells to pBC1-hLF-transfected cells were 1.29 and 1.59, respectively. hLF can promote spontaneous and irradiated H460 cells apoptosis and reduce the ability of colony formation (P <0.01). Conclusion High expression of hLF gene can significantly inhibit the growth of human non-small cell lung cancer H460 cells in vitro and enhance its radiation sensitivity.
