目的:研究苦参碱(matrine,Mat)对血管紧张素Ⅱ(AngⅡ)诱导人胚肺成纤维细胞(HFL-I)增殖和胶原合成的影响,探讨其抑制纤维化的机制。方法:以AngⅡ诱导HFL-I为研究对象,不同浓度的Mat作用48 h后,采用四氮唑盐(MTT)法检测Mat对HFL-I增殖的影响;羟脯氨酸测定HFL-I胶原合成量。结果:在一定范围内,苦参碱以浓度依赖性方式抑制AngⅡ诱导的HFL-I增殖和胶原合成(P<0.01)。结论:苦参碱通过抑制AngⅡ诱导的HFL-I增殖和胶原合成增加,从而发挥有效的抗纤维化作用。
Objective: To investigate the effect of matrine (Mat) on the proliferation and collagen synthesis of human embryonic lung fibroblasts (HFL-I) induced by angiotensin Ⅱ (Ang II) and to explore its mechanism of inhibiting fibrosis. Methods: HFL-I was induced by AngⅡ, and the effect of Mat on the proliferation of HFL-I was detected by MTT assay after 48 h of different concentrations of Mat. Hydroxyproline was used to determine the synthesis of HFL-I collagen the amount. Results: In a certain range, matrine inhibited AngⅡ-induced HFL-I proliferation and collagen synthesis in a concentration-dependent manner (P <0.01). Conclusion: Matrine can effectively inhibit the fibrosis by inhibiting the proliferation of HFL-I induced by AngⅡ and the increase of collagen synthesis.
