目的 :探讨丹参抗肝纤维化的可能机制。方法 :链蛋白酶、胶原酶离体灌注大鼠肝脏 ,Nycodenz密度梯度离心分离肝星状细胞 (HSC)体外培养。不同浓度的丹参处理 ,用RT -PCR法半定量检测HSC的转化生长因子 β1(TGF - β1)mRNA与Ⅰ型胶原mRNA的表达。结果 :丹参对HSC的TGF - β1与Ⅰ型胶原mRNA的表达有抑制作用。结论 :丹参抗肝纤维化作用可能与TGF - β1和Ⅰ型胶原mRNA的表达下调有关。
Objective: To explore the possible mechanism of salvia miltiorrhiza anti-fibrosis. METHODS: Rat livers were perfused with exogenous proteases and collagenase. Nycodenz density gradient centrifugation was used to isolate hepatic stellate cells (HSC) in vitro. Different concentration of Salvia miltiorrhiza was treated with semi-quantitative RT-PCR method to detect the expression of transforming growth factor β1 (TGF-β1) mRNA and type I collagen mRNA in HSC. Results: Salvia miltiorrhiza can inhibit the expression of TGF-β1 and type I collagen mRNA in HSC. Conclusion: The effect of Salvia miltiorrhiza on liver fibrosis may be related to the down-regulation of TGF-β1 and type I collagen mRNA expression.
