目的以威海沿海产多棘海盘车为原料,确定多糖的最佳提取工艺,并测定其抑菌活性。方法以苯酚-硫酸分光光度法测定多糖含量,考察酶解法,碱法,碱-酶结合等提取工艺的条件和优越性;采用滤纸片法测定海星多糖体外抑菌作用。结果氢氧化钠浓度为1mol·L-1,料液比为1∶2,提取2h时有最大产率。提取物对金黄色葡萄球菌有一定抑制作用。结论碱法优于其他制备方法;多棘海盘车粗多糖对细菌的抑制作用具有选择性。
Objective To determine the optimum extraction process of polysaccharid seaweeds from Weihai coast and determine its antibacterial activity. Methods The content of polysaccharides was determined by phenol-sulfuric acid spectrophotometry. The conditions and superiority of the extraction methods such as enzymatic hydrolysis, alkaline method and alkali-enzyme combination were investigated. The inhibitory effect of starfish polysaccharides in vitro was determined by filter paper method. Results The concentration of sodium hydroxide was 1mol·L-1, the ratio of solid to liquid was 1:2, and the maximum yield was obtained after 2h extraction. The extract has a certain inhibitory effect on Staphylococcus aureus. Conclusion Alkali method is superior to other preparation methods; polysaccharides of A. spinosa have selective effect on the inhibition of bacteria.
