据最近报告,9—氨基—1、2、3、4四氢吖啶(tetrahydroacridine,THA)同卵磷脂合用,治疗阿尔茨海默病有效.THA是一种胆硷酯酶抑制剂.也有认为是一种NMDA受体拮抗剂,除对胆硷酯酶的抑制作用之外,还可与毒蕈硷及烟硷受体发生作用,以减少乙酰胆硷的破坏及释放.阿尔茨海默病常表现有胆硷能活性的降低.在肌萎缩侧索硬化患者脊髓胆硷乙酰转移酶以及毒蕈硷的结合点明显减少.这表明,胆萎缩侧索硬化(ALS)亦存在胆硷能障碍.作者对8例诊断明确的ALS患者进行了试验性治疗.接受治疗者首先肌注30mg普鲁本辛(propanthelin)以减少副作用,45分钟后静脉注射30mgTHA,并记录各种检查指标.口服持续约7周,开始每隔日给药25mg,之后每日25mg,渐加量直至出现明显副作用,或达到最大剂量每日200mg时为止.每日用
