采用RT-PCR技术从橡胶树的叶片中分离到一条852 bp的c DNA,该序列包含一个798 bp的开放读码框(ORF),21 bp的5’UTR和33 bp的3’UTR;ORF预测编码265个氨基酸,理论分子量为28.64 k Da,等电点为5.55,属于叶绿体定位的类囊体膜蛋白;蛋白含有1个保守的叶绿素结合结构域,3个跨膜螺旋和2个C端螺旋,1个三聚化基序,以及多个叶绿素和类胡萝卜素结合位点,隶属于LHCB1亚家族;蛋白与蓖麻(Ricinus communis)、葡萄(Vitis vinifera)和棉花(Gossypium hirsutum)中同源蛋白的相似性都在95%以上,显示出高度的保守性,将基因命名为Hb Lhcb2.2。表达分析显示,Hb Lhcb2.2主要在叶片中表达,其表达水平随着叶片的成熟而逐渐增加,而在衰老叶片中则显著下调。
A 852 bp cDNA was isolated from the leaves of the rubber tree by RT-PCR. The sequence contained a 798 bp open reading frame (ORF), 21 bp 5’UTR and 33 bp 3’UTR. The protein contains a conserved chlorophyll-binding domain, three transmembrane helices and two C-terminal helices , One trimerization motif and several chlorophyll and carotenoid binding sites, belonging to the LHCB1 subfamily; proteins homologous to genes in Ricinus communis, Vitis vinifera and Gossypium hirsutum Protein similarity is above 95%, showing a high degree of conservation, the gene named Hb Lhcb2.2. Expression analysis showed that Hb Lhcb2.2 was mainly expressed in leaves, its expression gradually increased with the maturation of leaves, but significantly decreased in aged leaves.
