以5个有重要育种价值的籼型恢复系为材料,研究了影响籼稻愈伤组织诱导、生长和再生的几个重要因素。结果表明,对于籼稻品种而言,MS,NB培养基都是较为适合的培养基类型,愈伤诱导率都比较高,且愈伤质量较好,具有较强的再生能力,但是不同品种对不同培养基的反应有所不同,籼稻品种之间再生能力存在极大差异;分化培养基中适宜的激素配比为BAP3mg/L+NAA0.5mg/L;适当的干燥处理可以极大地提高愈伤组织的再生能力。还从这5个品种中初步确定了3个再生频率较高的品种,作为进一步研究籼稻组织培养特性的材料,并进行下一步的遗传转化试验。
Five important indica restorer lines with important breeding value were used as materials to study several important factors affecting the induction, growth and regeneration of indica rice callus. The results showed that for indica rice varieties, MS and NB mediums were all suitable medium types with higher callus induction rate, better callus quality and stronger regeneration ability, but different varieties were different The reaction of the medium was different, and the regenerative ability of indica rice varieties showed great difference. The appropriate hormone ratio in the differentiation medium was BAP3mg / L + NAA0.5mg / L. Appropriate drying treatment could greatly improve the callus Regenerative ability. Three varieties with higher regeneration frequency were also preliminarily identified from these five varieties as materials for further study on tissue culture characteristics of indica rice and the next genetic transformation test was conducted.
