目的阐述多重食源性致病菌核酸检测系统在一起副溶血性弧菌食物中毒事件检测中的运用。方法采用多重PCR方法对提取的DNA模板进行扩增,扩增后的产物进行全自动毛细管电泳,最后结合自动分析软件对毒力基因特异性条带进行结果判断。结果 37份样品中,检出5株副溶血性弧菌,均携带tdh、tlh和toxR基因,不携带trh基因。结论多重食源性致病菌核酸检测方法可以同时鉴定副溶血性弧菌及其携带毒力基因情况,具有检测快速、方便、自动化程度高等特点,此方法可在疾控机构监测食源性致病菌、食物中毒暴发事件中推广运用。
Objective To describe the application of multiple food-borne pathogenic bacteria nucleic acid detection system in the detection of food poisoning caused by Vibrio parahaemolyticus. Methods The multiplexed PCR method was used to amplify the extracted DNA template. The amplified product was subjected to full-automatic capillary electrophoresis. Finally, the results of the virulence gene-specific bands were determined by automatic analysis software. Results Among 37 samples, 5 strains of Vibrio parahaemolyticus were detected, all carrying tdh, tlh and toxR genes and no trh gene. Conclusion The nucleic acid detection method of multiple food-borne pathogens can identify Vibrio parahaemolyticus and its virulence genes at the same time. It has the characteristics of rapid, convenient and high degree of automation. This method can be used to monitor the food-induced Germs, food poisoning outbreaks in the promotion and application.
