本文以逆转录-DNA聚合酶链反应和U_(937)细胞cDNA文库的分离杂交方法研究了肺纤维化患者支气管肺泡洗出液(BAL)细胞的基因表达情况。结果表明,在大部分标本可检出白细胞介素-1β和肿瘤坏死因子αmRNA,在部分标本中可检出转化生长因子β和胰岛素样生长因子-I mRNA;肺纤维化患者的BAL细胞可表达出一些正常志愿者BAL细胞未表达的mRNA,而这些mRNA的拷贝存在于PMA刺激的U_(937)细胞cDNA文库中。
In this paper, the gene expression of bronchoalveolar lavage fluid (BAL) in patients with pulmonary fibrosis was studied by reverse transcription-DNA polymerase chain reaction and U937 cell cDNA library hybridization. The results showed that IL-1β and TNFα mRNA were detected in most of the specimens, and TGF-β and insulin-like growth factor-I mRNA were detected in some specimens. BAL cells in patients with pulmonary fibrosis were expressed Some normal volunteers did not express BAL cells mRNA, and copies of these mRNAs were present in the PMA-stimulated U 937 cell cDNA library.
