目的 :探讨碱性成纤维细胞生长因子 (b FGF)对庆大霉素 (GM)诱导的急性肾功能衰竭 (ARF)的疗效。方法 :雄性 Wistar大鼠 32只 ,随机分为对照组、实验组 (GM组 )中的 GM1组、GM2组、GM3组各 8只。GM组按 16 0 mg/kg.d- 1肌注 GM8d,对照组注射等体积生理盐水。于实验第 9天从对照组和 GM1组动物腹主动脉取血样做血清尿素氮 (BU N)和血清肌酐 (Scr)测定后 ,经心脏灌注固定 ,取肾皮质做光镜检查。剩下的 GM2组、GM3组动物于实验第 9天停止肌注 GM,GM2组按 b FGF2 0 0 0 U/kg.d- 1 肌注 8d,GM3组则注射等体积的生理盐水 ,之后又于第 9天从 GM2组、GM3组动物中取血样做生化检查、取肾皮质做光镜和电镜观察。结果 :b FGF可促进 GM诱导的损伤肾小管上皮细胞的恢复和再生 ,但血清学变化不明显。结论 :1b FGF对 GM诱导的 ARF有一定的治疗效果 ;2 Scr和 BU N的正常并不代表肾脏无病理改变
Objective: To investigate the effect of basic fibroblast growth factor (b FGF) on gentamicin-induced acute renal failure (ARF). Methods: Thirty-two male Wistar rats were randomly divided into control group, experimental group (GM group), GM1 group, GM2 group and GM3 group. The GM group was injected intramuscularly with GMO at 16 mg / kg.d-1 for 16 days, and the control group was injected with equal volume of normal saline. Blood samples were taken from the abdominal aorta of the control group and the GM1 group on day 9 of the experiment to measure the blood urea nitrogen (BU N) and serum creatinine (Scr), and then fixed by heart perfusion. The renal cortex was examined by light microscopy. The remaining GM2 and GM3 animals stopped intramuscular injection of GM on the 9th day of the experiment, the GM2 group received intramuscular injection of bFGF20 000 U / kg.d-1 for 8 days, and the GM3 group received an equal volume of normal saline, Blood samples were taken from the GM2 group and the GM3 group on the 9th day for biochemical examination and renal cortex for light and electron microscopy. RESULTS: b FGF promoted the recovery and regeneration of injured renal tubular epithelial cells induced by GM, but the serological changes were not obvious. Conclusion: 1b FGF has a certain therapeutic effect on GM-induced ARF. The normal expression of 2 Scr and BU N does not mean that the kidney has no pathological changes
