目的:探讨藤莪清瘀方对人胃癌BGC-823细胞株的抑制及凋亡诱导作用。方法:采用不同浓度的藤莪清瘀方提取物(25.00、12.50、6.25、3.12、1.25mg/m L)体外处理人胃癌BGC-823细胞24、48、72h;采用四甲基偶氮唑蓝(MTT)法观察藤梨根不同浓度和不同作用时间对人胃癌BGC-823细胞增殖的抑制率;采用Annexin VFITC/PI双染法检测细胞凋亡情况。结果:藤莪清瘀方提取物原液药物浓度(50mg/m L)经稀释8倍(6.25mg/m L)后,抑制率最强(68.67±3.50)%,随着药物浓度的降低,抑制率随之下降;藤莪清瘀方对胃癌细胞作用48h抑制作用最强。藤莪清瘀方提取物处理细胞48h后,凋亡比率达33.15%。结论:藤莪清瘀方对胃癌细胞有明显抑制作用,并诱导其凋亡。
OBJECTIVE: To investigate the inhibitory effect of TZEQQ on human gastric cancer cell line BGC-823 and the induction of apoptosis. Methods: Human gastric cancer cell line BGC-823 was treated with different concentrations of Tengcu Qingyu Decoction (25.00,12.50,6.25,3.12,1.25mg / m L) for 24, 48 and 72 hours in vitro. Tetramethylbenzothiazolyl tetrazolium (MTT) method was used to observe the inhibitory rate of proliferation and proliferation of human gastric cancer cell line BGC-823 at different concentrations and different time periods. Cell apoptosis was detected by Annexin VFITC / PI double staining. Results: The best inhibition rate (68.67 ± 3.50)% was obtained after the concentration of drug solution (50mg / m L) was 8 times (6.25mg / m L) The rate of which decreased. The effect of TZEQQY on the gastric cancer cells 48h was the strongest. After 48 hours of treatment, the ratio of apoptosis was 33.15%. Conclusion: Fuju Qingyu Decoction significantly inhibited gastric cancer cells and induced its apoptosis.
