近几年,原位肝移植(OLT)已超越实验阶段用于临床。目前它已被公认为治疗一系列终末期肝病值得选择的方法。但OLT有许多不便,因此仍在继续寻找别的治疗措施,至少在某些情况下可作为它的替代疗法。其中肝细胞移植(HT)似乎是最优最有前途的方法之一,它已被证明对实验性急慢性肝衰有效。因而人们认为它可能作为暴发性肝炎和肝硬化的暂时性肝支持,作为先天性代谢性肝病的永久性肝支持。然而要把HT真正地用于人类,各种问题有待澄清,包括全面了解移植后肝细胞的功能和保存细胞的可靠方法。本文旨在评估移植冷冻一融化的肝细胞建立能保持肝细胞特性并最终成为肝支持的异位肝的可能性。
In recent years, orthotopic liver transplantation (OLT) has surpassed the experimental stage for clinical use. It has now been widely accepted as a treatment of choice for a range of end-stage liver diseases. However, the OLT has many inconveniences and is therefore continuing to search for alternative treatments, at least in some cases as an alternative. Among them, hepatocyte transplantation (HT) seems to be one of the best and most promising methods, and it has been shown to be effective in experimental acute and chronic liver failure. It is therefore considered as a possible temporary liver support for fulminant hepatitis and cirrhosis as a permanent liver support for congenital metabolic liver disease. However, HT needs to be truly used in humans and various issues remain to be clarified, including a thorough understanding of the function of hepatocytes after transplantation and a reliable method of preserving cells. This article aims to evaluate the possibility of transplantation of frozen-thawed hepatocytes to establish an ectopic liver that retains hepatocyte properties and ultimately becomes liver support.
