本文从还原剂SnCl_3用量,反应体系pH值,McAb使用浓度等方面研究了~(99m)Tc直接法标记单克隆抗体的最佳条件,并探讨了VitC对标记反应的影响。β-ME法还原单抗,三氯乙酸沉淀法测定标记率,上行纸层析法测定标记产物胶体含量。结果表明,(99m)Tc直接标记中还原剂SnCl_2浓度以0.01~0.04mg/ml为宜,反应最适pH值在5.6~6.8,McAb最低使用浓度应为500μg/ml以上,适量的VitC对标记反应有促进作用。~(99m)Tc-2F_7McAb在荷瘤裸鼠的肾、肝、脾、肿瘤中分布较高,其它器官内放射性相对较低。瘤/血比在24hr后可达2.0左右,有一定的诊断应用价值。
In this paper, the optimal condition for the direct labeling of monoclonal antibody by ~(99m)Tc was studied from the aspects of the amount of reducing agent SnCl_3, the pH value of the reaction system, and the concentration of McAb. The effect of VitC on the labeling reaction was also discussed. The monoclonal antibody was reduced by β-ME method and the labeling rate was determined by trichloroacetic acid precipitation method. The colloid content of the labeled product was determined by up-paper chromatography. The results showed that the concentration of reducing agent SnCl 2 in the (99m)Tc direct labeling was 0.01-0.04mg/ml, the optimum pH was 5.6-6.8, the minimum concentration of McAb was 500μg/ml, and the appropriate amount of vitamin C was labeled The reaction has a promoting effect. The ~(99m)Tc-2F_7McAb was highly distributed in the kidney, liver, spleen and tumor of tumor-bearing nude mice, and the radioactivity in other organs was relatively low. The tumor/blood ratio can reach 2.0 or so after 24 hours, which has certain diagnostic application value.
