目的研究多发性骨髓瘤(MM)患者外周血单个核细胞(PBMC)中c-Cbl和Cbl-b基因表达情况。方法收集23例MM患者和22例健康人PBMC,利用SYBRGreen实时荧光定量PCR方法检测MM患者和健康人PBMC中c-Cbl和Cbl-b基因相对表达水平;利用RT-PCR方法分别扩增MM患者和健康人c-Cbl基因第7~10外显子,并对PCR产物进行序列分析。结果 MM组c-Cbl表达水平(中位数0.798%),与健康对照组c-Cbl表达水平(中位数2.443%)相比,显著降低(P<0.05);Cbl-b表达水平(中位数0.714%)与健康对照组Cbl-b表达水平(中位数2.179%)比较,也显著降低(P<0.05﹚。在2例MM患者中检测到c-Cbl基因第7~10外显子存在2个大小不同的片段:483 bp和148 bp,分别为野生型c-Cbl和删除突变的c-Cbl基因,所有PCR产物测序后均未发现突变。结论 MM患者PBMC中c-Cbl和Cbl-b基因表达下调。
Objective To investigate the expression of c-Cbl and Cbl-b in peripheral blood mononuclear cells (PBMCs) from patients with multiple myeloma (MM). Methods The PBMCs of 23 patients with MM and 22 healthy controls were collected. The relative expression levels of c-Cbl and Cbl-b genes in PBMC from patients with MM and healthy controls were detected by SYBRGreen real-time PCR. The relative expression levels of c-Cbl and Cbl-b were detected by RT- MM patients and healthy people c-Cbl gene exons 7 to 10, and PCR products were sequenced. Results The expression level of cbl-b in MM group was significantly lower than that in healthy control group (median 0.798%, median 2.443%, P <0.05) The median of 0.714%) was significantly lower than that of healthy control group (median 2.179%) (P <0.05). In 2 MM patients, there were two fragments of different sizes in exon 7 to exon 10 of c-Cbl gene: 483 bp and 148 bp, which were wild-type c-Cbl and deleted mutant c-Cbl, all No mutations were found after PCR products were sequenced. Conclusion The expression of c-Cbl and Cbl-b in PBMC of MM patients is down-regulated.
