目的探讨孕早期膳食对子代肥胖和肥胖相关基因解偶联蛋白3(UCP3)的影响,寻求合理的孕早期膳食和早期防治肥胖的靶标。方法 Wistar孕鼠随机分为两组,孕早期分别食用标准膳食(SD)和高蛋白膳食(HPD),孕中、晚期两组均食用SD,所产雄性子鼠分别为对照(CON)组和高蛋白(HP)组,3周断乳后食用SD至成年,将CON组的大鼠随机分为两组:一组继续食用SD(CON组);另一组高脂膳食诱导肥胖,为高脂对照(HFC)组,处死大鼠取材,检测UCP3mRNA的表达。结果与对照组相比,HP组大鼠的体重降低(P<0.05);UCP3的表达持续升高。结论孕早期HPD可通过程序性升高子代UCP3基因的表达,降低子代成年肥胖的发生。
OBJECTIVE: To investigate the effects of early pregnancy diet on obesity-related genes uncoupling protein 3 (UCP3) in the first trimester of pregnancy and to find a reasonable diet for early pregnancy and the target of early prevention and treatment of obesity. Methods Wistar pregnant rats were randomly divided into two groups. SD and HPD were consumed during the first trimester of pregnancy. SD was fed to the second and third trimester of both groups. The male mice were control group (CON) and control group High-protein (HP) group. After 3 weeks of weaning, SD was induced to infancy, and rats in CON group were randomly divided into two groups: one group continued to consume SD (CON group); the other high-fat diet induced obesity The rats in the HFC group were sacrificed and the expression of UCP3 mRNA was detected. Results Compared with the control group, the body weight of rats in HP group was decreased (P <0.05), and the expression of UCP3 was continuously increased. Conclusion HPD during early pregnancy may reduce the incidence of adult obesity in children by increasing the expression of UCP3 gene.
