目的应用多位点序列分型(multi-locus sequence typing,MLST)技术对副溶血性弧菌进行基因分型,探讨菌株间的内在联系与同源性,分析其优势型。方法收集2006-2012年,宁波市食物中毒和散发腹泻患者副溶血性弧菌48株,用生化鉴定确定其表型;用MLST方法对菌株进行基因分型。结果 48株副溶血性弧菌分出9个ST型和1个未定型,其中ST3 32株(66.67%);ST265 5株(10.42%);ST1203株(6.25%);ST8 2株(4.17%);ST262 1株(2.08%);ST199 1株(2.08%);ST332 1株(2.08%);ST527 1株(2.08%);ST189 1株(2.08%);未定型1株(2.08%)。1株副溶血性弧菌的等位基因分别为dnaE234、gyrB4、recA162、dtdS222、pntA、pyrC184和tnaA79,与MLST数据库中无匹配,为1个国内新的副溶血性弧菌ST型(ST262)。结论宁波市副溶血性弧菌流行株以ST3克隆群为主。首次从国内副溶血性弧菌中分离出ST262型,有进一步分析研究意义。MLST分型能发现副溶血性弧菌间的关联,并与国内外菌株进行比较,发现新的ST型,是一种较好的分型方法。
Objective To genotype Vibrio parahaemolyticus using multi-locus sequence typing (MLST) technique to explore the intrinsic relationship and homology among isolates. Methods 48 strains of Vibrio parahaemolyticus were collected from food poisoning and diarrhea patients in Ningbo City from 2006 to 2012, and their phenotypes were determined by biochemical identification. The strains were genotyped by MLST. Results 48 strains of Vibrio parahaemolyticus were divided into 9 ST types and 1 unconfined type, of which ST3 32 strains (66.67%), ST265 5 strains (10.42%), ST1203 strains (6.25%), ST8 2 strains ); ST262 1 strain (2.08%); ST199 strain 1 (2.08%); ST332 strain 1 (2.08%); ST527 strain 1 (2.08% . One allele of Vibrio parahaemolyticus was dnaE234, gyrB4, recA162, dtdS222, pntA, pyrC184 and tnaA79, respectively. There was no match with MLST database, which was a new ST strain (ST262) . Conclusion Ningbo epidemic strains of Vibrio parahaemolyticus ST3 clonal group based. For the first time isolated from domestic Vibrio parahaemolyticus ST262 type, further analysis of the significance. MLST typing can be found in the correlation between Vibrio parahaemolyticus, and compared with domestic and foreign strains, found that the new ST type, is a better typing method.
