目的研究IL-23在体内外对肝细胞肝癌发展的影响。方法采用慢病毒体系将构建好的IL-23质粒转入Hepa1-6肝癌细胞株中,流式分选得到过表达IL-23的Hepa1-6稳转细胞,ELISA确定所得到的稳转细胞能够表达高水平的IL-23。采用MTT方法检测IL-23对Hepa1-6细胞增殖的影响,流式细胞染色法检测IL-23对Hepa1-6细胞周期的影响,通过原位种植Hepa1-6细胞建立肝癌模型,检测IL-23体内在肝细胞肝癌发展中的作用。结果 IL-23能够明显促进肝癌细胞的增殖,然而对肝癌细胞的凋亡却没有影响,IL-23在体外促进肝癌细胞的迁移水平,并且在小鼠肝癌模型中,IL-23在体内能够促进肝癌的发展。结论IL-23在体内体外能够直接促进肝细胞肝癌的发展。
Objective To study the effect of IL-23 on the development of hepatocellular carcinoma in vitro and in vivo. Methods The constructed IL-23 plasmid was transfected into Hepa1-6 hepatocarcinoma cell line by lentivirus system, and Hepa1-6 stable transfectant cells overexpressing IL-23 were obtained by flow cytometry. The obtained stable transfected cells could be determined by ELISA Express high levels of IL-23. The effects of IL-23 on the proliferation of Hepa1-6 cells were detected by MTT assay. The effects of IL-23 on the cell cycle of Hepa1-6 cells were detected by flow cytometry. The Hepa1-6 cells were inoculated with HepG1 cells in situ to detect the expression of IL-23 Role of the body in the development of hepatocellular carcinoma. Results IL-23 could significantly promote the proliferation of hepatoma cells, but had no effect on the apoptosis of hepatoma cells. IL-23 promoted the migration of hepatocellular carcinoma cells in vitro, and IL-23 promoted the in vivo Liver cancer development. Conclusion IL-23 can directly promote the development of hepatocellular carcinoma in vitro and in vivo.
