目的:观察TGF-β1在阻塞性黄疸肝组织中的表达,并初步探讨其作用。方法:运用Wistar大鼠建立阻寨性黄疸模型,观察其肝脏病理变化及血清胆红素的变化;运用免疫组织化学方法检测肝脏组织中TCF-β1的表达;并运用原位末端标记方法检测肝细胞凋亡。结果:Wistar大鼠结扎胆总管后,胆红素急剧升高,术后1周可见肝脏表达TCF-β1,肝细胞出现大量的凋亡,纤维组织增生,术后2周TGF-β1表达及细胞凋亡达到高峰,纤维组织大量增生,术后3周纤维组织占据肝脏组织大部分,细胞凋亡相对减少,仍可见大量的TGF-β1表达。结论:在阻塞性黄疸中,肝脏表达大量的TGF-β1,与肝细胞凋亡、纤维化及机体免疫功能抑制密切相关。
Objective: To observe the expression of TGF-β1 in the liver tissue of obstructive jaundice and to explore its role. Methods: Wistar rats were used to establish the model of obstructive jaundice. The changes of hepatic pathological changes and serum bilirubin were observed. The expression of TCF-β1 in liver tissues was detected by immunohistochemical method. The expression of TCF- Apoptosis. Results: Bilirubin was significantly increased in the Wistar rats after ligation of the common bile duct. The expression of TCF-β1 was observed in the liver one week after operation, with significant apoptosis of hepatocytes, fibrosis, and the expression of TGF-β1 Apoptosis reached a peak, a large number of fibrous tissue hyperplasia, 3 weeks after the fibrous tissue occupy most of the liver tissue, the relative reduction of apoptosis, still visible a large number of TGF-β1 expression. CONCLUSION: In obstructive jaundice, the liver expresses a large amount of TGF-β1, which is closely related to the inhibition of hepatocyte apoptosis, fibrosis and immune function.
