目的观察6-羟多巴(6-hydroxydopamine,6-OHDA)损伤大鼠黑质损伤后下丘脑室旁核(paraventricular nucleus of hypothalamus,PVN)中离子钙接头蛋白1(ionized calcium binding adapter molecule-1,Iba1)的含量变化。方法 30只SD大鼠根据随机数字表分为6-OHDA大鼠组和对照组各15只。6-OHDA大鼠组双侧黑质内注射6-羟多巴,对照组双侧黑质(substantia nigra,SN)内注射等体积的生理盐水。6周后断头取脑,Nissl染色、免疫组化、免疫印迹方法鉴定成模,免疫组化、免疫印迹和RT-PCR检测室旁核Iba1的表达。结果与对照组比较,6-OHDA大鼠组SN内酪氨酸羟化酶(tyrosine hydroxylase,TH)阳性神经细胞数量从(58±5)个减少到(10±2)个(P<0.05),蛋白表达水平从(0.71±0.12)降低至(0.28±0.12)(P<0.05),室旁核中Iba1阳性细胞数量从(7±2)个增加到(14±3)个(P<0.05),蛋白表达水平从(0.1±0.08)增加到(0.19±0.06)(P<0.05)。结论 6-OHDA大鼠PVN中Iba1的表达变化有可能与帕金森病患者的胃肠功能障碍存在一定的联系。
Objective To observe the effects of ionized calcium binding adapter molecule-1 (I-1) in paraventricular nucleus of hypothalamus (PVN) after 6-hydroxydopamine (6-OHDA) , Iba1) content changes. Methods Thirty SD rats were divided into 6-OHDA group and control group according to random number table. 6-OHDA rats were injected with 6-OHDA into the substantia nigra and substantia nigra (SN) of the control group. Six weeks later, brain was cut off, Nissl staining, immunohistochemistry and immunoblotting were used to identify the expression of Iba1 in paraventricular nucleus. Immunohistochemistry, Western blotting and RT-PCR were used to detect the expression of Iba1. Results Compared with the control group, the number of tyrosine hydroxylase (TH) positive neurons in the SN of 6-OHDA group decreased from 58 ± 5 to 10 ± 2 (P <0.05) (P <0.05). The number of Iba1 positive cells in paraventricular nucleus increased from (7 ± 2) to (14 ± 3), P <0.05 ), The protein expression level increased from (0.1 ± 0.08) to (0.19 ± 0.06) (P <0.05). Conclusion The change of Iba1 expression in PVN of 6-OHDA rats may be related to the gastrointestinal dysfunction in patients with Parkinson’s disease.
