目的:分析黄芪注射液对宫颈癌细胞的促增殖效应及其相关调控分子表达的影响。方法:将人宫颈癌Hela细胞与系列稀释后终浓度分别为生药7.82 g/L、15.63g/L、31.25g/L、62.50g/L、125g/L、250g/L、500g/L、1000g/L、2000g/L、4000g/L(相当于每升原药材克数)的黄芪注射液共同孵育,噻唑蓝(MTT)法检测细胞生长增殖的A492值,流式细胞计数(FCM)分析细胞周期各时相细胞百分率及凋亡指数(AI)、增殖指数(PI);免疫细胞化学法检测细胞中PCNA、Cyclin A、Bcl-2、Bax等增殖凋亡相关调控分子表达的变化,计算表达阳性细胞百分率,并以图像分析系统定量分析阳性表达强度。结果:生药125g/L~4000g/L的黄芪注射液作用后,可使Hela细胞生长增殖的A492值呈递进性显著增高;Hela细胞G0/G1期比例及AI值显著降低,G2/M期比例及PI值显著升高,S期比例未见显著差异。同时,Hela细胞中PCNA阳性表达率、Cyclin A及Bcl-2阳性表达率及表达强度明显增高,Bax的阳性表达率及表达强度明显降低。结论:黄芪注射液可通过影响Hela细胞增殖凋亡相关调控分子的表达而发挥促进生长增殖的作用。黄芪制剂应用于宫颈癌患者及具此类肿瘤诱发体质人群的其他疾病治疗中须慎用。

Conclusion: Astragalus injection can play a role in promoting growth and proliferation by affecting the expression of apoptosis-related regulatory molecules in Hela cells. Astragalus preparations should be used with caution in the treatment of patients with cervical cancer and other diseases induced by such tumors.
