目的 探讨日本血吸虫组织蛋白酶BDNA疫苗 (Sj31B1N)抗生殖免疫的可能机理。 方法 用Sj31B1N免疫小鼠 ,每 2周 1次 ,每次免疫前均尾静脉采血 1次 ,作血清抗体动态变化分析 ,免疫 4次后尾蚴攻击感染。感染后 4 5天 ,计数成虫负荷和肝、肠组织内虫卵数。结果 用Sj31B1N免疫小鼠后第 2周部分小鼠血清中出现了抗体 ,随时间延长 ,出现抗体阳性组小鼠数量增多 ,抗体滴度增加。抗体阳性组小鼠与阴性组小鼠对比 ,成虫减少率为 4 0 .6% ,肝组织减卵率为 4 8.0 % ,肠组织减卵率为 4 5.4 %。结论 核酸疫苗Sj31B1N可有效地诱导小鼠产生抗体 ,并且具有免疫保护作用。核酸疫苗Sj31B1N所诱导的体液免疫可能是抗日本血吸虫生殖免疫的机理之一。
Objective To investigate the possible mechanism of schistosoma japonicum cathepsin B DNA vaccine (Sj31B1N) in anti-reproductive immunity. Methods The mice were immunized with Sj31B1N once every two weeks, and the blood samples were collected one time before each immunization. The serum antibodies were analyzed for the dynamic changes. After four immunizations, the cercariae challenged the infection. 45 days after infection, the number of adult worms and the numbers of eggs in the liver and intestine tissues were counted. Results Antibody was detected in the serum of mice in the second week after immunization with Sj31B1N. With the passage of time, the number of antibody positive mice increased and the antibody titer increased. Compared with the negative group, the rate of adult reduction was 40.6%, the rate of egg reduction was 4 8.0%, and the rate of egg reduction was 44.4%. Conclusion The nucleic acid vaccine Sj31B1N can effectively induce antibody production in mice and has immunoprotective effect. Humoral immunity induced by nucleic acid vaccine Sj31B1N may be one of the mechanisms of anti-Schistosoma japonicum reproductive immunity.
