目的探讨表没食子儿茶素没食子酸酯(EGCG)是否具有协同塞来昔布抑制鼻咽癌细胞CNE-2增殖及下调环氧化酶-2(COX-2)表达。方法体外培养CNE-2细胞,与EGCG或(和)塞来昔布孵育24~72 h,MTT法检测细胞增殖情况,流式细胞术分析细胞周期,Western blot检测COX-2的表达。结果 EGCG对塞来昔布抑制CNE-2细胞增殖具有协同作用,能协同增加G0~G1期细胞比例、降低S、G2~M期细胞数,促进细胞凋亡,且能协同协调COX-2的表达。结论 EGCG对COX-2抑制剂的协同效应可做为鼻咽癌化疗潜在的辅助用药。
Objective To investigate whether epigallocatechin-3-gallate (EGCG) has a synergistic effect on the proliferation of nasopharyngeal carcinoma cell line CNE-2 and the expression of cyclooxygenase-2 (COX-2). Methods CNE-2 cells were cultured in vitro and incubated with EGCG or celecoxib for 24-72 h. Cell proliferation was measured by MTT assay. Cell cycle was analyzed by flow cytometry. COX-2 expression was detected by Western blot. Results EGCG had a synergistic effect on the proliferation of CNE-2 cells treated with celecoxib, which could synergistically increase the proportion of cells in G0-G1 phase, decrease the number of cells in S phase and G2-M phase and promote cell apoptosis, and could coordinate the COX-2 expression. Conclusion The synergistic effect of EGCG on COX-2 inhibitors could be used as a potential adjuvant for the treatment of nasopharyngeal carcinoma.
