凝溶胶蛋白(Gelsolin)作为一种重要的肌动蛋白调节蛋白,在机体的多种生理和病理活动中发挥重要作用。本文采用细胞MTT染色法,Hoechst33342/PⅠ荧光染色和流式细胞仪技术,对浙江枝吻纽虫(Dendrorhynchus zhejiangensis)的gelsolin基因的原核表达产物(r-gelsolin)进行了体外抗肿瘤活性的研究。结果表明,30μg/ml的r-gelsolin作用于胃低分化粘液腺癌MGC803细胞24h后,细胞的增殖抑制率达到最高,约为34.53%。荧光染色显示,r-gelsolin使MGC803细胞的正常形态发生改变,细胞核出现核固缩,破裂,产生凋亡小体等现象。进一步通过流式细胞仪证实,r-gelsolin可以阻滞MGC803细胞于S、G2期,影响细胞分裂的进程,并促使细胞发生凋亡。
Gelsolin, as an important actin regulatory protein, plays an important role in many physiological and pathological activities of the body. In this study, MTT assay, Hoechst33342 / PⅠ fluorescent staining and flow cytometry were used to study the antitumor activity of gelsolin in Zhejiang Dendrobium zhejiangensis. The results showed that 30μg / ml r-gelsolin could inhibit the proliferation of gastric mucosal adenocarcinoma MGC803 cells for 24 h, and the highest inhibitory rate reached 34.53%. Fluorescence staining showed that r-gelsolin changed the normal morphology of MGC803 cells, and nuclear pyknosis, rupture and apoptotic bodies appeared. Further confirmed by flow cytometry, r-gelsolin MGC803 cells can block the S, G2 phase, affecting the process of cell division, and promote apoptosis of cells.
