目的:探讨盐酸川芎嗪联合不同比例的富血小板血浆(platelet-rich plasma,PRP)对人骨髓间充质干细胞(Bone Marrow Mesenchymal Stem Cells,BMSCs)体外增殖及其对成骨细胞分化的影响。方法:抽取已签订知情同意书的患者静脉血50 m L,二次离心法制备PRP,分别进行血小板计数分析。将体积分数为15%、30%、50%的PRP与80%盐酸川芎嗪按照1∶1.5混合加入骨髓间充质干细胞的培养液中,细胞达到80%融合,消化收集细胞,检测细胞的增殖状况;根据计数确定各组细胞培养,观察PRP和盐酸川芎嗪组对细胞培养的影响,并对PRP和盐酸川芎嗪组培养的细胞进行成骨诱导,确定其成骨分化特性。结果:各组PRP+盐酸川芎嗪促能够进MSCs增殖,达到80%融合时间更短,MSCs增殖明显优于对照组;MSCs增殖与PRP浓度呈正相关关系,随着PRP浓度增加细胞增殖效率显著增加。结论:PRP与盐酸川芎嗪对人骨髓间充质干细胞的体外增殖有促进作用,与PRP体积分数和作用时间呈正相关关系。
Objective: To investigate the effects of ligustrazine hydrochloride combined with platelet-rich plasma (PRP) on proliferation and differentiation of human bone marrow mesenchymal stem cells (BMSCs) in vitro. Methods: The venous blood of patients with signed informed consent was drawn for 50 m L and PRP was prepared by secondary centrifugation. Platelet count analysis was performed respectively. The volume fraction of 15%, 30%, 50% PRP and 80% ligustrazine hydrochloride were mixed according to 1: 1.5 mixed with bone marrow mesenchymal stem cells in culture medium, the cells reached 80% confluency, digestion and collection of cells, detection of cell proliferation The cell culture of each group was determined by counting. The effects of PRP and ligustrazine hydrochloride on cell culture were observed. The osteogenic differentiation of cells cultured with PRP and ligustrazine hydrochloride was induced by osteogenic differentiation. Results: The effect of PRP + Ligustrazine Hydrochloride on the proliferation of MSCs was better than that of the control group. The proliferation of MSCs was positively correlated with the concentration of PRP. The proliferation of MSCs increased significantly with the increase of PRP concentration. Conclusion: PRP and ligustrazine hydrochloride can promote the proliferation of human bone marrow mesenchymal stem cells in vitro, and have a positive correlation with the volume fraction of PRP and the action time.
