目的 观察缺氧诱导因子 - 1α在慢性脑缺血大鼠海马中的表达 ,并探讨其表达的意义。方法 采用永久性双侧颈总动脉结扎术 ( 2 VO)制备大鼠慢性脑缺血的动物模型 ,运用 Western印迹和免疫组化检测 HIF- 1α的表达。结果 Western印迹显示 2 VO1d后 HIF- 1α的表达即上调 ,2周后进一步升高 ,保持这一水平直至第 6周。免疫组化见海马各区均有免疫阳性细胞分布。结论 HIF- 1α在慢性脑缺血大鼠海马中的表达上调 ,可能参与了慢性脑缺血的代偿机制。
Objective To observe the expression of hypoxia inducible factor - 1α in hippocampus of rats with chronic cerebral ischemia and to explore its significance. Methods The rat model of chronic cerebral ischemia was established by permanent bilateral common carotid artery ligation (2 VO). The expression of HIF-1α was detected by Western blotting and immunohistochemistry. Results Western blotting showed that the expression of HIF-1α was up-regulated after 2 VO1d and further increased after 2 weeks, maintaining this level up to the 6th week. Immunohistochemistry in the hippocampus all have immunoreactive cells distribution. Conclusion HIF-1α is up-regulated in hippocampus of rats with chronic cerebral ischemia and may be involved in the compensatory mechanism of chronic cerebral ischemia.
