目的研究补骨脂素对A375细胞黑素合成的影响以及丝裂原活化蛋白激酶(MAPK)信号通路的作用。方法将补骨脂素作用于A375细胞,采用MTT法检测细胞活性;Na OH裂解法检测黑素量;多巴氧化法检测酪氨酸酶(TYR)活性;RT-PCR法测定TYR、酪氨酸酶相关蛋白-1和酪氨酸酶相关蛋白-2(TRP-1、TRP-2)以及ERK1、ERK2、JNK2这3个MAPK信号通路关键蛋白激酶的m RNA表达。结果补骨脂素在安全剂量下可显著抑制A375细胞黑素合成及TYR活性,1滋mol·L-1补骨脂素能明显下调A375细胞TYR、TRP-1、TRP-2以及ERK1、ERK2、JNK2 m RNA表达。结论推测补骨脂素通过抑制ERK1、ERK2、JNK2的表达来抑制TYR活性和/或下调TYR、TRP-1及TRP-2 m RNA表达,抑制A375细胞的黑素合成。
Objective To study the effect of psoralen on the melanogenesis of A375 cells and the role of mitogen-activated protein kinase (MAPK) signaling pathway. Methods Psoralen was applied to A375 cells. The cell viability was measured by MTT assay. The content of melanin was detected by Na OH cleavage method. Tyrosinase (TYR) activity was detected by dopa oxidation assay. The tyrosinase activity TRP-1, TRP-2, ERK1, ERK2 and JNK2, which are key MAPKs in MAPK signaling pathway. Results Psoralen could significantly inhibit the melanin synthesis and TYR activity of A375 cells at a safe dosage. 1 p mol·L-1 psoralen significantly reduced TYR, TRP-1, TRP-2 and ERK1, ERK2 , JNK2 m RNA expression. Conclusions Psoralen suppresses TYR, TRP-1 and TRP-2 mRNA expression and inhibits melanogenesis in A375 cells by inhibiting the expression of ERK1, ERK2 and JNK2.
