目的探讨脂肪源性干细胞(ADSC)移植对大鼠脑缺血再灌注损伤区炎症递质多聚腺苷酸二磷酸核糖聚合酶(PARP)和核因子-κB(NF-κB)表达的影响。方法采用线栓法复制大鼠大脑中动脉缺血再灌注模型,将动物随机分为假手术组、模型组和ADSC组。ADSC组经尾静脉注射PKH-26标记的ADSC细胞悬液(0.5ml,细胞密度为4×106个/ml),观察各组大鼠的神经功能损害评分,采用蛋白质印迹法和PCR分别检测半暗带区NF-κB和PARP蛋白及其mRNA的表达水平。结果 ADSC移植后可见PKH26标记的ADSC在半暗带区有表达。与模型组比较,ADSC组神经功能损害评分显著降低(P<0.05),半暗带区PARP蛋白、NF-κB蛋白和mRNA表达水平均显著下降(P<0.05)。结论脂肪源性干细胞移植可通过下调炎症递质NF-κB和PARP的表达抑制炎性反应,从而促进脑缺血损伤神经功能的恢复。
Objective To investigate the effects of adipose-derived stem cells (ADSCs) transplantation on the expression of inflammatory mediators poly (ADP-ribose) polymerase (PARP) and nuclear factor-κB (NF-κB) in rats with focal cerebral ischemia-reperfusion injury. Methods The rat middle cerebral artery occlusion (MCAO) model was reproduced by thread occlusion. Animals were randomly divided into sham operation group, model group and ADSC group. ADSC group were injected with PKH-26 labeled ADSC cell suspension (0.5ml, cell density 4 × 106 cells / ml) through the tail vein to observe the neurological impairment scores of rats in each group. Western blotting and PCR were used to detect the half Dark zone of NF-κB and PARP protein and mRNA expression levels. Results After ADSC transplantation, PKH26-labeled ADSCs were expressed in the penumbra zone. Compared with the model group, the scores of neurological impairment in ADSC group were significantly decreased (P <0.05), and the expression of PARP protein, NF-κB protein and mRNA in penumbra zone were significantly decreased (P <0.05). Conclusion Adipose-derived stem cell transplantation can inhibit the inflammatory response by down-regulating the expression of inflammatory mediators NF-κB and PARP, thereby promoting the recovery of neurological function after cerebral ischemia injury.
