代谢控制分析(MetabolicControlAnalysis,简称MCA)是一种以系统观点对细胞内代谢调控进行定量分析的理论和实验方法。根据MCA理论,代谢控制是细胞内代谢的系统属性,并可以用酶动力学性质进行定量表示。MCA指出了“限速步骤”观念的局限性,认为代谢途径中酶促反应步骤对代谢的控制作用随系统内外条件的改变而变化。用理论计算或试验确定的控制系数能够加深对代谢的控制结构特性的理解。MCA与计算机仿真技术和各种现代生物实验技术有机结合,正成为代谢工程中越来越重要的手段和一种有效的研究模式。目前MCA还处在发展完善之中,需要扩大其研究和应用的范围,积累更多的应用实例
Metabolic Control Analysis (MCA) is a theoretical and experimental method for the quantitative analysis of intracellular metabolic regulation from a systemic point of view. According to the MCA theory, metabolic control is a systemic property of intracellular metabolism and can be quantified using enzymatic properties. MCA points out the limitations of the concept of “rate-limiting steps,” arguing that the metabolic control of the enzymatic steps in the metabolic pathway varies with changes in the system’s internal and external conditions. Using theoretical calculations or experimentally determined control factors can deepen understanding of the metabolic control structural properties. The combination of MCA with computer simulation technology and various modern biological experiment technologies is becoming more and more important means and an effective research mode in metabolic engineering. At present, MCA is still in perfect development and needs to expand the scope of its research and application and accumulate more application examples
