目的观察王氏连朴饮对脾胃湿热证模型大鼠胃黏膜增殖与凋亡相关P53、Bcl-2和COX-2蛋白表达的影响,探讨该方治疗脾胃湿热证获效的可能作用机制。方法取SD大鼠50只,随机分为正常对照组,模型组,王氏连朴饮高、中、低剂量组(1.94,0.97,0.48 g·m L-1),共5组,每组10只。除正常组饲以普通饲料外,其余4组均以湿热环境加高脂高糖饮食和白酒综合法复制脾胃湿热证模型,连续灌胃给药7 d,观察该方对模型大鼠胃黏膜P53、Bcl-2及COX-2蛋白表达的影响。结果本实验P53、Bcl-2、COX-2基因蛋白表达定位于胞质,模型组大鼠表达明显升高,与正常对照组比较,有显著性差异(P<0.01)。王氏连朴饮高、中、低剂量组P53、Bcl-2及COX-2蛋白表达有一定程度下调,与模型组比较,具有显著性差异(P<0.01),尤以中剂量组作用最明显。结论王氏连朴饮可能通过下调脾胃湿热模型大鼠胃黏膜P53、Bcl-2及COX-2蛋白水平的表达,在一定程度上调节其胃黏膜细胞增殖与凋亡的失衡,进而达到对胃黏膜损伤的修复作用。
Objective To observe the effect of Wang Lian Pu Yin on the expression of P53, Bcl-2 and COX-2 protein in gastric mucosa of rats with spleen-stomach damp-heat syndrome and to explore the possible mechanism of this prescription in treating damp-heat syndrome of spleen and stomach. The expression of P53, Bcl-2 and COX-2 in the model group was significantly higher than that in the normal control group (P <0.01). The expression of P53, Bcl-2 and COX-2 in Wanglianpu high, medium and low dose groups were down-regulated to a certain extent, which was significantly different from the model group (P <0.01) obvious. Conclusions Wang Lian Pu Yin may regulate the expression of P53, Bcl-2 and COX-2 protein in gastric mucosa of spleen-stomach damp-heat model rats to a certain extent, regulate the imbalance between the proliferation and apoptosis of gastric mucosa cells, Mucosal injury repair effect.
