目的为探讨山楂叶总黄酮(HLF,w/w,80%flavonoids)抗血栓的潜能,研究了山楂叶总黄酮对缺氧人脐静脉内皮细胞的调控作用。方法应用流式细胞技术,检测了内皮细胞细胞毒及胞内一氧化氮水平。此外,运用激光共焦点扫描显微技术,检测了细胞内钙离子水平。结果研究数据显示,5μg/ml和10μg/ml的山楂叶总黄酮可以降低缺氧人脐静脉内皮细胞的细胞毒水平(P<0.05,P<0.01)。缺氧内皮细胞内钙离子和一氧化氮的水平也能被5μg/ml(P<0.01,P<0.01)和10μg/ml(P<0.01,P<0.01)的山楂叶总黄酮显著下调。结论结果提示,山楂叶总黄酮对缺氧人脐静脉内皮细胞有保护作用,且下调细胞内钙离子和一氧化氮的水平可能是这种保护作用的机制之一。
Objective To investigate the antithrombotic potential of hawthorn leaves flavonoids (HLF, w/w, 80% flavonoids) and to study the regulation of total flavonoids from hawthorn leaves on hypoxia human umbilical vein endothelial cells. Methods Flow cytometry was used to detect endothelial cell cytotoxicity and intracellular nitric oxide levels. In addition, intracellular calcium levels were measured using laser confocal scanning microscopy. Results The data showed that total flavonoids of Hawthorn leaves at 5 and 10 μg/ml reduced the cytotoxicity of human umbilical vein endothelial cells (P<0.05, P<0.01). The levels of calcium and nitric oxide in hypoxic endothelial cells were also significantly down-regulated by total flavonoids in Hawthorn leaves at 5 μg/ml (P<0.01, P<0.01) and 10 μg/ml (P<0.01, P<0.01). Conclusion The results suggest that hawthorn leaf flavonoids have protective effects on hypoxia human umbilical vein endothelial cells, and downregulation of intracellular calcium and nitric oxide levels may be one of the mechanisms of this protective effect.
