目的:通过建立多囊卵巢综合征(PCOS)高雄血症动物模型,观察金雀异黄素(GEN)对大鼠子宫和卵巢形态学的影响。方法:按照胰岛素(INS)联合HCG造模法建立实验动物模型,分为阴性对照组、模型组、GEN低、中、高剂量组及雌激素阳性对照组,剂量组分别灌胃给予GEN5、10、20mg/kgbw,雌激素组给予己烯雌酚0.5mg/kg,持续15d,观察动情周期10d。试验结束后,取大鼠各脏器称重,计算脏体比值、卵巢和子宫的面积与体积。显微镜下分析子宫和卵巢组织形态学的变化。结果:与模型组相比,中、高剂量组大鼠的子宫/体比升高,差异有显著性(p<0.05);各剂量组大鼠卵巢面积和体积均下降,雌激素组下降明显,有显著性差异(p<0.05)。卵巢大体和镜下观察结果显示,高剂量GEN可使造模后大鼠的卵巢卵泡膜细胞层变薄,颗粒细胞层数明显增多,形态完整,排列整齐,黄体组织数量增多,成熟卵泡居多,囊肿卵泡明显减少。结论:GEN对PCOS高雄血症大鼠的子宫和卵巢形态学起到了改善作用,推测GEN的植物雌激素样作用可能会对PCOS患者发挥治疗作用。

Ovarian general and microscopic observations showed that high dose of GEN can make the rat ovarian follicular cell layer thinning, granulosa cells significantly increased the number of layers, complete morphology, neatly arranged, the number of luteal tissue increased, the majority of mature follicles, Cyst follicles significantly reduced. Conclusion: GEN can improve the morphology of uterus and ovary in rats with PCOS, and it is speculated that the phytoestrogen-like effect of GEN may play a therapeutic role in patients with PCOS.
