目的:建立测定人血浆中曲唑酮浓度的高效液相色谱法。方法:以DiamonsilTM C18反相柱(150mm×4.6mm,5μm)为色谱柱,流动相为0.03mol.L-1醋酸铵-甲醇(24∶76);流速:0.8mL·min-1;柱温:40℃;检测波长:255nm。以乙酸乙酯与二氯甲烷(80∶20)为提取剂。结果:曲唑酮的高、中、低(960.0,120.0,10.0ng·mL-1)3种浓度平均回收率分别为97.26%,98.42%,101.26%,日内、日间差RSD均低于7%(n=5);分析方法的检测限为5.0ng·mL-1;线性范围为10.0~960.0ng.mL-1。曲线方程:Y=0.36X+2.12,r=0.9992(n=9)。结论:该方法灵敏、准确、简单、快速,可用于临床血药浓度监测和药动学研究。
Objective: To establish a HPLC method for the determination of trazodone in human plasma. METHODS: DiamonsilTM C18 reversed-phase column (150mm × 4.6mm, 5μm) was used as the mobile phase with a mobile phase of 0.03mol·L-1 ammonium acetate-methanol (24:76), flow rate of 0.8mL · min- : 40 ° C; detection wavelength: 255 nm. Ethyl acetate and dichloromethane (80:20) as extractant. Results: The mean recoveries of trazodone at high, medium and low concentrations (960.0,120.0,10.0 ng · mL -1) were 97.26%, 98.42% and 101.26%, respectively. The intra-day and inter-day RSDs were lower than 7 % (n = 5). The detection limit of the analytical method was 5.0 ng · mL-1. The linear range was 10.0-960.0 ng.mL-1. Curve equation: Y = 0.36X + 2.12, r = 0.9992 (n = 9). Conclusion: The method is sensitive, accurate, simple and rapid, which can be used for clinical blood drug concentration monitoring and pharmacokinetic studies.
