目的研究小鼠卵母细胞减数分裂恢复过程中Akt1和Akt2在从GV期向GVBD期转化期间亚细胞定位的特点。方法采用间接免疫分析技术观察Akt1和Akt2在小鼠卵母细胞GV期和GVBD期的定位;利用显微注射实验技术将Akt1和Akt2抗体分别注射入卵细胞,在不同时间点观察GVBD的发生的数量。结果 Akt1在小鼠卵母细胞GV期均匀的分布于细胞质,而在GVBD期则分布于细胞膜。Akt2在GV期和GVBD期分布于整个细胞,但是GV期以细胞质更为密集,而在GVBD期以细胞核附近更为密集。结论 Akt1可能参与小鼠卵母细胞减数分裂恢复的调控。
Objective To study the characteristics of subcellular localization of Akt1 and Akt2 during GVBD phase transition from meiotic resumption of mouse oocyte meiosis. Methods Indirect immunoassay was used to observe the location of Akt1 and Akt2 in GV stage and GVBD stage of mouse oocytes. Akt1 and Akt2 antibodies were injected into the oocytes respectively by microinjection, and the number of GVBD was observed at different time points . Results Akt1 was evenly distributed in the cytoplasm of mouse oocytes during GV phase and distributed in the plasma membrane during GVBD phase. Akt2 is distributed throughout the GV and GVBD phases, but is more cytoplasmic in the GV phase and closer to the nucleus in the GVBD phase. Conclusion Akt1 may be involved in the regulation of mouse oocyte meiosis recovery.
