目的:研究亚砷酸(As2O3)联合顺铂(DDP)对人鼻咽癌CNE-2Z裸鼠移植瘤的生长抑制作用及对死亡相关蛋白激酶(DAPK)的影响。方法:建立人鼻咽癌裸鼠移植瘤模型,随机分为对照组、As2O3组、DDP组和As2O3加DDP组,比较各组的抑瘤作用;并对标本分别进行光镜、原位末端标记(TUNEL)检测,采用实时荧光定量PCR法与免疫组织化学方法检测DAPK的表达。结果:As2O3和As2O3加DDP均能显著抑制人鼻咽癌裸鼠移植瘤的生长,诱导肿瘤细胞凋亡,并能上调DAPK的表达。结论:As2O3可抑制人鼻咽癌CNE-2Z裸鼠移植瘤的生长,该作用与诱导人鼻咽癌CNE-2Z裸鼠移植瘤细胞凋亡、上调DAPK的表达相关。As2O3与DDP联用可起协同作用。
OBJECTIVE: To investigate the effects of arsenious acid (As2O3) and cisplatin (DDP) on the growth of human nasopharyngeal carcinoma CNE-2Z xenografted in nude mice and its effect on the death-associated protein kinase (DAPK). Methods: The nude mice model of human nasopharyngeal carcinoma was established and randomly divided into control group, As2O3 group, DDP group and As2O3 plus DDP group. The antitumor effect of each group was compared. The specimens were examined by light microscopy, in situ end-labeling (TUNEL) assay. The expression of DAPK was detected by real-time fluorescence quantitative PCR and immunohistochemistry. Results: Both As2O3 and As2O3 plus DDP could significantly inhibit the growth of human nasopharyngeal carcinoma xenografts in nude mice, induce tumor cell apoptosis and up-regulate the expression of DAPK. CONCLUSION: As2O3 can inhibit the growth of human nasopharyngeal carcinoma CNE-2Z xenografts in nude mice, which may be related to the induction of apoptosis and the up-regulation of DAPK expression in human nasopharyngeal carcinoma CNE-2Z xenografts. As2O3 and DDP can play a synergistic effect.
