目的:探讨血红素加氧酶-1(HO-1)在慢性肾衰竭大鼠肾脏中的表达、活性及其诱导剂对肾脏病变的保护作用。方法:将大鼠随机分为三组:A组假手术组,B组慢性肾衰竭组,C组给予氯化高铁血红素(Hemin)灌饲慢性肾衰竭组,8周后检测大鼠血压,肾功能,血、尿蛋白,观察肾组织病理改变,免疫组织化学观察HO-1的表达。结果:C组较B组蛋白尿水平明显降低(P<0.05),肾功能明显降低(P<0.05),但较A组明显升高(P<0.05),肾小球硬化及肾小球基底膜增生程度以及肾间质纤维化程度虽较B组减轻,但差异无显著性意义。免疫组织化学显示,B组HO-1的表达较A组、C组均明显降低(平均光度、面密度)(P<0.05),但肾组织中B组、C组之间HO的活性无统计学意义,B、C组较A组均明显降低(P<0.05),血清中HO的活性B组较C组明显降低(P<0.05)。结论:HO-1在慢性肾衰竭大鼠肾脏中的表达及活性是降低的,但其诱导剂对慢性肾衰竭大鼠肾脏病变具有部分保护作用。其机制尚可待进一步研究。
Objective: To investigate the expression and activity of heme oxygenase-1 (HO-1) in the kidney of rats with chronic renal failure and the protective effect of its inducer on renal lesions. Methods: The rats were randomly divided into three groups: sham operation group A, B group chronic renal failure group, C group were given hemin for chronic renal failure group, 8 weeks later the blood pressure, Renal function, blood and urine protein were observed. The pathological changes of kidney were observed and the expression of HO-1 was observed by immunohistochemistry. Results: The proteinuria in group C was significantly lower than that in group B (P <0.05) and the renal function was significantly decreased (P <0.05), but significantly higher than that in group A (P <0.05). Glomerular sclerosis and glomerular basement Although the degree of membrane hyperplasia and the degree of renal interstitial fibrosis were lower than those in group B, the difference was not significant. Immunohistochemistry showed that the expression of HO-1 in group B was significantly lower than that in group A and group C (mean luminosity and areal density) (P <0.05), but the activity of HO in group B and C was not statistically significant In group B and C, the activity of HO in group B was significantly lower than that in group A (P <0.05). CONCLUSION: The expression and activity of HO-1 in the kidney of chronic renal failure rats is decreased, but its inducer has a partial protective effect on renal lesions in rats with chronic renal failure. Its mechanism can be further studied.
