目的 研究大黄对烫伤加内毒素“二次打击”后大鼠肝细胞线粒体内单价氧自由基泄漏的影响。方法 采用大鼠背部烫伤模型 ,烫伤 12h后给予内毒素 ( 2 0mg/kg)“二次打击” ,监测肝细胞线粒体内细胞色素氧化酶的活力 ,同时测定肝细胞线粒体内过氧化物歧化酶 (SOD)活力。结果 脓毒症大鼠肝细胞线粒体内细胞色素氧化酶的活力随时间而降低 ,大黄能显著提高细胞色素氧化酶的活力。脓毒症大鼠肝细胞线粒体内SOD大量消耗 ,而大黄能显著降低肝细胞线粒体内SOD的消耗。结论 大黄能显著提高脓毒症大鼠肝细胞线粒体内SOD和细胞色素氧化酶的活力 ,降低肝细胞线粒体内单价泄漏。
Objective To study the effect of rhubarb on mitochondrial oxygen free radical leakage in rat hepatocytes after scald plus endotoxin “second strike”. Methods Rat model of scald back injury was established. Endotoxin (20 mg / kg) was administered twice a day after scald for 12 hours. The activity of mitochondrial cytochrome oxidase in liver cells was monitored. The activities of mitochondrial superoxide dismutase SOD) vitality. Results The activity of mitochondrial cytochrome oxidase in liver cells of septic rats decreased with time, and rhubarb could significantly increase the activity of cytochrome oxidase. The sepsis rats liver cells mitochondrial SOD consumption, and rhubarb can significantly reduce the liver mitochondrial SOD consumption. Conclusion Rhubarb can significantly increase the activity of mitochondrial SOD and cytochrome oxidase in liver cells of septic rats, and reduce monochromatic leakage in liver cells.
