目的 应用流式细胞术(FCM)对60例大肠癌组织的S期细胞百分比(SPF)进行检测;应用免疫组化方法对60例大肠癌p16基因产物表达进行检测,探讨p16表达与细胞增殖活性之间的关系。方法 应用流式细胞仪FACCS Calibur,激发波长488um,DNA染色试剂为PI进行检测,分析采用Modfit LT2.0软件。结果 p16表达阴性者的SPF明显高于p16表达阳性者的SPF,两者间具有极显著差异。结论 p16失表达、失活间接促进了大肠癌细胞增殖。p16表达水平可以作为检测大肠癌增殖活性的指标。
Objective To detect the percentage of S phase cells (SPF) in 60 cases of colorectal cancer by flow cytometry (FCM); to detect the expression of p16 gene product in 60 cases of colorectal cancer by immunohistochemistry to explore p16 expression and cell proliferation activity. The relationship between. Methods Flow cytometer FACCS Calibur was used. The excitation wavelength was 488um. The DNA staining reagent was detected by PI. The analysis was performed using Modfit LT2.0 software. Results The SPF of p16-negative patients was significantly higher than that of p16-positive patients. There was a significant difference between the two groups. Conclusion The loss of expression and inactivation of p16 indirectly promote the proliferation of colorectal cancer cells. The expression level of p16 can be used as an index for detecting the proliferative activity of colorectal cancer.
