目的探讨葡萄籽提取物原花青素对卵巢癌SKOV3细胞增殖和凋亡的影响和机制。方法体外培养的SKOV3细胞与25、50、100、200、400μg/ml的葡萄籽提取物原花青素共孵育,分别在24、48、72h时观察细胞形态学变化,MTT检测细胞增殖抑制率,流式细胞和TUNEL分析细胞凋亡,RT-PCR检测survivin在mRNA的表达情况,Western blot检测SKOV3细胞survivin在蛋白水平的表达。结果不同浓度(25、50、100、200、400μg/ml)原花青素对SKOV3细胞作用48h的细胞增殖抑制率分别为48.67%、53.85%、67.03%、73.78%、77.39%。流式细胞和TUNEL检测经25μg/ml原花青素处理24、48h后SKOV3细胞凋亡率分别为31.98%、45.78%。葡萄籽原花青素还可以明显降低survivin的mRNA和蛋白水平的表达。这些作用均随原花青素浓度和作用时间的延长而增强。结论原花青素可能通过降低survivin的表达,在体外抑制SKOV3细胞增殖,并促进其凋亡。
Objective To investigate the effect and mechanism of grape seed extract procyanidin on proliferation and apoptosis of ovarian cancer SKOV3 cells. Methods SKOV3 cells cultured in vitro were incubated with proanthocyanidins (25, 50, 100, 200 and 400 μg / ml). The morphological changes of cells were observed at 24, 48 and 72 hours respectively. Cell apoptosis was analyzed by TUNEL. Survivin mRNA was detected by RT-PCR and Western blot was used to detect the protein expression of survivin in SKOV3 cells. Results The proliferation inhibition rate of SKOV3 cells treated with proanthocyanidins at different concentrations (25,50,100,200,400μg / ml) for 48h were 48.67%, 53.85%, 67.03%, 73.78% and 77.39%, respectively. Flow cytometry and TUNEL detection of 25μg / ml proanthocyanidins treated 24,48h after the apoptosis rate of SKOV3 cells were 31.98%, 45.78%. Grape seed proanthocyanidins can also significantly reduce the expression of survivin mRNA and protein levels. These effects are enhanced with the proanthocyanidins concentration and duration of action. Conclusion Proanthocyanidins may inhibit the proliferation and induce the apoptosis of SKOV3 cells in vitro by decreasing the expression of survivin.
