在体外染毒条件下,观察了不同浓度的二硫化碳(CS2)对大鼠神经细胞内游离钙离子浓度([Ca2+]i)、蛋白激酶C(PKC)活性和培养的神经细胞骨架的影响。结果显示,在一定浓度范围内,CS2引起[Ca2+]i和PKC活性超生理水平的升高,并呈现剂量-反应关系,且[Ca2+]i和PKC活性的变化具有一致性;随着CS2浓度的增加,大鼠神经细胞骨架网络的破坏程度发生由轻到重的变化,也表现为剂量-反应关系。实验结果提示CS2引起神经细胞内信号传递系统的异常变化,导致细胞结构的破坏,可能是CS2产生神经毒性的重要原因。
The effects of different concentrations of carbon disulfide (CS2) on the intracellular free calcium concentration ([Ca2 +] i), protein kinase C (PKC) activity and neuronal cytoskeleton were observed in vitro. The results showed that at a certain concentration range, CS2 caused a hyper-physiological increase of [Ca2 +] i and PKC activity, and showed a dose-response relationship. The change of [Ca2 +] i and PKC activity was consistent. , The severity of neuronal cytoskeleton network changes from light to heavy, also showed a dose-response relationship. The experimental results suggest that CS2 causes abnormal changes of the signal transduction system in nerve cells, leading to the destruction of cell structure, which may be an important reason for the neurotoxicity of CS2.
