目的:分离前列腺癌组织中细胞分化抑制蛋白1(Id1)的相互结合蛋白,以探讨Id1在前列腺癌中的作用途径和机制。方法:首先构建PolyHis-Id1的表达载体pET-28a/Id1,并在大肠埃希菌BL21中进行诱导表达,然后采用牵出试验钓取前列腺癌组织中的Id1相互结合蛋白。结果:蛋白电泳并染色后发现一清晰蛋白条带,用活化转录因子3抗体经Western印迹尝试检测发现其确为活化转录因子3。结论:在人前列腺癌组织中,Id1可能与活化转录因子3相互结合发挥效应。
OBJECTIVE: To isolate the interaction protein of cell differentiation inhibitor 1 (Id1) in prostate cancer tissue and to explore the role of Id1 in prostate cancer. Methods: The expression vector pET-28a / Id1 of PolyHis-Id1 was constructed and induced in Escherichia coli BL21. The Id1-binding protein in prostate cancer tissues was extracted by pull-out test. Results: A clear protein band was found after electrophoresis and staining of the protein, and it was found to be activated transcription factor 3 by Western blotting using the activated transcription factor 3 antibody. Conclusion: In human prostate cancer, Id1 may interact with activated transcription factor 3 to exert its effect.
