蛋白免疫是传统的抗体制备法,基因免疫是新型的抗体制备法。为了制备抗P16抗血清以及比较基因免疫和蛋白免疫制备抗体,分别以融合蛋白谷胱甘肽S—转移酶-P16(GST-P16)和克隆有p16肿瘤抑制基因相关cDNA的裸露真核表达载体pCMV-p16经皮内多点注射法接种家兔,制备抗P16抗血清,Western印迹法检测到蛋白免疫的抗P16抗血清滴度为1∶625,明显高于基因免疫制备的抗P16抗血清滴度(1∶5)。说明了基因免疫普及于制备抗体前,还需对其特点和加强免疫反应的方法深入研究
Protein immunization is a traditional antibody preparation method. Gene immunization is a new antibody preparation method. In order to prepare anti-P16 antiserum and compare the immunogenicity and immunogenicity of antibodies, we constructed a recombinant eukaryotic expression vector containing the fusion protein glutathione S-transferase-P16 (GST-P16) and the cDNA cloned with the p16 tumor suppressor gene pCMV-p16 was inoculated intradermally in rabbits to prepare anti-P16 antiserum. The titer of protein-immunized anti-P16 antiserum was 1: 625, which was significantly higher than that of gene-immunized anti-P16 antiserum Titer (1: 5).
