目的 :探讨丙型肝炎 (丙肝 )患者外周血单个核细胞 (PBMCs)内丙型肝炎病毒抗原 (HCVAg)及Fas抗原 (FasAg)表达状况及意义。方法 :采用HCV核心区及NS3 区单克隆抗体 ,对 2 8例慢性丙肝患者PBMCs进行免疫组化检测 ,并同步检测FasAg。结果 :HCVAg及FasAg阳性率分别为 85 7% (2 4/ 2 8)和 82 2 % (2 3/ 2 8) ,经相关分析显示。两者表达呈显著正相关 (r =0 5 46 ,P <0 0 1)。FasAg阳性PBMCs中的部分细胞有凋亡性改变。结论 :HCV可感染PBMCs,并可激发FasAg表达。PBMCs中的HCV和Fas介导的细胞凋亡 ,会影响机体免疫功能 ,并可能成为病毒持续感染的原因之一。
Objective: To investigate the expression and significance of hepatitis C virus antigen (HCVAg) and Fas antigen in peripheral blood mononuclear cells (PBMCs) of patients with hepatitis C (HCV). Methods: PBMCs from 28 chronic hepatitis C patients were detected by immunohistochemistry using HCV core region and NS3 monoclonal antibody, and FasAg was detected simultaneously. Results: The positive rates of HCVAg and FasAg were 85 7% (2 4/2 8) and 82 2% (2 3/2 8), respectively. There was a significant positive correlation between them (r = 0546, P <0.01). Some of FasAg-positive PBMCs have apoptotic changes. Conclusion: HCV can infect PBMCs and induce FasAg expression. HCV and Fas-mediated apoptosis in PBMCs can affect the immune function of the body and may be one of the causes of persistent virus infection.
