目的探讨固醇调节元件结合蛋白1c(SREBP-1c)、激素敏感脂肪酶(HSL)和脂肪组织甘油三酯脂肪酶(ATGL)在不同体重指数(BMI)人群的皮下脂肪组织(SAT)和大网膜脂肪组织(OAT)中的mRNA表达。方法按BMI将82例患者分为超重组(A组,35例,BMI 24-27.9)、肥胖组(B组,27例,BMI≥28)和正常体重组(C组,20例,BMI 18.5-23.9)。检测三组血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL)、高密度脂蛋白胆固醇(HDL);半定量RT-PCR方法检测三组腹部SAT和OAT中SREBP-1c、HSL、ATGL 3个基因的mRNA表达水平。结果A、B组的血脂异常率高于C组(42.86%、51.86%vs.10.00%)(P<0.05)。在SAT中,B组SREBP-1cmRNA表达高于C组(P<0.05);A、B组的HSL和ATGL mRNA表达水平低于C组(P<0.05)。在OAT中,A、B组的SREBP-1c、ATGL mRNA表达高于C组,而HSL mRNA表达则低于C组(P<0.05)。结论超重和肥胖人群SAT和OAT的脂代谢基因表达水平紊乱,可能是导致其血脂异常率升高、更易罹患内分泌代谢及心血管疾病的机制之一。
Objective To investigate the effects of steroid regulatory element binding protein 1c (SREBP-1c), hormone-sensitive lipase (HSL) and adipose tissue triglyceride lipase (ATGL) on subcutaneous adipose tissue MRNA expression in omental adipose tissue (OAT). Methods According to BMI, 82 patients were divided into three groups: overweight group (A group, 35 cases, BMI 24-27.9), obesity group (B group, 27 cases, BMI≥28) and normal weight group (C group, 20 cases, BMI 18.5 -23.9). The levels of total cholesterol (TC), triglyceride (TG), low density lipoprotein cholesterol (LDL) and high density lipoprotein cholesterol (HDL) were measured in the three groups. -1c, HSL, ATGL mRNA expression levels of three genes. Results The rates of dyslipidemia in group A and group B were higher than those in group C (42.86%, 51.86% vs.10.00%, P <0.05). In SAT, the expression of SREBP-1 mRNA in group B was higher than that in group C (P <0.05). The mRNA expression of HSL and ATGL in group A and B was lower than that in group C (P <0.05). In OAT, the mRNA expression of SREBP-1c and ATGL in group A and group B was higher than that in group C, while the expression of HSL mRNA was lower in group A and B than that in group C (P <0.05). Conclusions The expression of lipid metabolism genes in overweight and obese subjects is disorderly, which may be one of the mechanisms leading to the increase of dyslipidemia and the more susceptible to endocrine and cardiovascular diseases.
