目的:观察癌基因Stat3、CyclinD1及增殖细胞核抗原(PCNA)在非小细胞肺癌(NSCLC)中的表达情况,并分析其在非小细胞肺癌发生发展中的作用。方法:应用免疫组织化学SP法检测45例NSCLC中 Stat3蛋白、CyclinD1蛋白及PCNA蛋白的表达。结果:45例NSCLC中32例Stat3蛋白阳性表达,阳性表达率为71.1%;NSCLC不同组织学分级、有无淋巴结转移Stat3的表达差异有显著性(P<0.05),肿瘤不同大小及不同组织学分型间表达差异无显著性(P>0.05);Stat3与CyclinD1蛋白及PCNA蛋白的表达呈正相关(r= 0.531,P<0.01;r=0.618,P<0.01)。结论:Stat3在NSCLC中异常表达,并可能参与NSCLC中CyclinD1蛋白的调节,促进细胞增殖,Stat3可能促进NSCLC侵袭和转移。
Objective: To observe the expression of oncogene Stat3, CyclinD1 and proliferating cell nuclear antigen (PCNA) in non-small cell lung cancer (NSCLC) and to analyze its role in the development of non-small cell lung cancer. Methods: The expressions of Stat3, CyclinD1 and PCNA in 45 cases of NSCLC were detected by immunohistochemical SP method. Results: Stat3 protein was positive in 32 of 45 NSCLC samples, the positive rate was 71.1%. The expression of Stat3 in different histological grade and lymph node metastasis of NSCLC was significantly different (P <0.05) (P> 0.05). There was a positive correlation between Stat3 and CyclinD1 protein and PCNA protein expression (r = 0.531, P <0.01; r = 0.618, P <0.01). CONCLUSIONS: Stat3 is abnormally expressed in NSCLC and may be involved in the regulation of CyclinD1 protein in NSCLC to promote cell proliferation. Stat3 may promote the invasion and metastasis of NSCLC.
